Description:
Phage display of various cDNA libraries on filamentous phage have resulted in the isolation of a vast number of cloned oligopeptides, whole proteins or domains thereof with specificity for target molecules or cells.

For antibodies, these techniques allow the isolation of monoclonal antibodies of any species. The antibodies are initially produced as fragment molecules (Fab or scFv) in bacteria, but isolated genes can be modified for eukaryotic expression as whole IgG. Primarily, the techniques facilitate the isolation of antibodies to antigens previously very difficult to raise antibodies to, and generation of monoclonal antibodies from other species than mice, e.g. humans, rabbits etc. Peptide libraries, consisting of randomised linear or cyclised oligopeptides expressed on phage, allow the isolation of peptide ligands to the target of choice. Peptide libraries can also be constructed to carry a certain conserved motif combined with randomised regions.

Infrastructure/methods:
We have a "know-how but no-hands" facility service. This means that we will assist in the selection of wanted antibodies or peptides from phage libraries under the following conditions:
• A person from the customers group will work in our lab for a limited time (6-10 weeks).
• Randomised peptide and antibody libraries are available for immediate screening; we will assist in the construction of new ones (this will need additional time).
• The selection procedure, and the subsequent expression of single clones, will be performed by the visiting person with our guidance.

Practical information:
Prior knowledge of molecular biology and recombinant expression techniques is an advantage and will facilitate the work.
